Targeted profiling with CETSA supports assay development in intact cells, enables scalable screening designs, and provides direct evidence of target engagement under physiologically relevant conditions. The approach can help identify novel chemotypes, differentiate mechanisms among compounds with similar potency, and highlight cases where functional effects do not align with enzymatic inhibition or expected binding modes. 

The method is therefore helpful when validating mechanistic hypotheses and confirming that on-target engagement occurs in the intended cellular context.
