Proteome-wide CETSA® profiling is a mass spectrometry-based method measuring compound-induced changes across thousands of proteins simultaneously. This enables detection of both direct target engagement and indirect pathway responses, providing a broad and physiologically relevant view of compound activity. 

This broad coverage helps uncover the molecular basis of compound activity, clarifies mechanisms that drive phenotypic outcomes, and identifies potential off-target effects that isolated biochemical assays often miss. Proteome-wide data can also support structure–activity exploration by showing how chemical changes influence engagement or pathway signatures.  

The depth and relevance of the data make proteome-wide CETSA ideal when the target is unknown, when multiple pathways may contribute to activity, or when early mechanistic insight is needed to support progression decisions.
