Bridge findings from cell systems to tissues and whole blood

Translational studies assess whether relationships observed in cellular models are maintained in relevant tissues and more complex biological systems. This phase focuses on linking exposure to pharmacodynamic effect, determining which matrices and time points are informative, and evaluating whether assay endpoints remain practical and interpretable across systems. The goal is to establish evidence that supports dose selection, sampling schedules, and endpoint choices for subsequent pharmacology and safety studies.

Study design decisions center on selecting tissues and matrices that best reflect the indication, such as whole blood, specific organs, or surrogate matrices. Sampling strategies are defined to capture relevant dose levels and time points, while analytical methods are selected to relate exposure to biological effects. Attention is given to sources of variability, including donor differences, tissue handling, and matrix interference, to ensure that observed effects are robust and reproducible.

What you see in cells, you are likely to see in patients

In translational studies, CETSA can be applied to assess target engagement directly in tissues or complex biological systems. This helps determine whether exposure–response relationships observed in cells extend to more physiologically relevant contexts and supports decisions on which endpoints are ready to progress into in vivo studies or biomarker verification.
